Knockout Inducible RNAi Systems
The Knockout Inducible RNAi Systems allow you to tightly regulate the expression of functional small hairpin RNAs (shRNAs) in mammalian cells for the purpose of silencing target genes. Knockout Inducible RNAi Systems are very useful in cases where suppression of a gene may be lethal, preventing its analysis. Basal expression of shRNA in the absence of induction is extremely low, which is crucial because even low expression levels of shRNA can result in significant suppression of the target gene. Additionally, our Inducible RNAi Systems provide high shRNA induction, ensuring strong protein knockdown.
There are three versions available.
The Knockout Single Vector Inducible RNAi system: once your shRNA is cloned this vector contains all the necessary features for inducible knockdown on a single plasmid. This system can save weeks of time by creating a stable inducible cell clone in a single step.
The Knockout Tet RNAi System H and Knockout Tet RNAi System P: These systems feature inducible retroviral shRNA expression vectors, which are provided pre-linearized and ready for ligation with a dsDNA oligonucleotide encoding an shRNA. The procedure is a 2 step process, creating of a tet responsive cell line by selection and screening of tTS stable cell clones, followed by selection and screening of clones that inducibly express your shRNA. To create the Tet responsive cell lines, you can either use the regulatory vectors provided in our inducible RNAi system to develop your own cell line specific to your interest, or you can start from one of our premade tTS stable regulatory cell lines and introduce your shRNA-response plasmid construct.
