Description

Lenti-X™ Tet-On® Advanced and Lenti-X Tet-Off® Advanced enable our award winning inducible gene expression systems to be established in the broadest variety of cell lines, nondividing cells, stem cells and tissues. Each system consists of:

- A regulatory vector that stably expresses either the Tet-On or Tet-Off Advanced transcriptional activator
- A response vector (pLVX-Tight-Puro) into which you clone your gene of interest
- Lenti-X HT Packaging System for highest possible titers
- Tet Approved Fetal Bovine Serum (FBS)
- A control response vector for luciferase expression

The expression level can be completely controlled by the level of Dox supplied in the medium. You can easily titrate the expression level, whether you wish to completely turn off, express to physiological levels, or over-express your protein.

It is possible to optimize the induction characteristics of cells transduced with these inducible Lenti-X Systems to meet specific expression needs. Obtain maximum expression of your gene of interest by infecting target cells with supernatant ratios that favor the response vector over the corresponding regulator vector. Alternatively, achieve maximum-fold induction and ensure the absolute lowest basal expression levels with supernatant ratios that favor the regulator vector.
