T3 DNA Ligase
T3 DNA Ligase catalyzes the formation of a phosphodiester bond between a 5′ phosphate and a 3′ hydroxyl terminus in double-stranded DNA.
The T3 DNA Ligase enzyme will ligate both sticky and blunt end termini, as well as repair single stranded nicks in dsDNA. Without the addition of 20-30% PEG 6000, T3 DNA Ligase has a very low blunt-end ligation efficiency. T3 DNA Ligase displays a higher ligation efficiency of A/T overhangs compared to C/G overhangs.
The enzyme is tolarant of moderate salt concentrations: T3 DNA Ligase retains 95% of its activity in 1.0 M NaCl or KCl, with an optimal concentration of 300 mM.
