The CRISPR/Cas9 Gesicles
The Guide-it CRISPR/Cas9 Gesicle Production System has been developed to obtain efficient delivery of the Cas9/sgRNA complex to all mammalian cell types with less off-target effects.
With this complete system, you can make cell-derived nanovesicles, the gesicles, that contain active Cas9 protein complexed with a sgRNA specific to your gene of interest.
The gesicles are produced by transfection of 293T cells; the formed gesicles end up in the medium, can be collected and immediately used to incubate target cells or stored at -70C for up to one year. 1. Gesicles are produced by 293T cells 2. Cas9/sgRNA complexes are enriched into the gesicles through the iDimerize system 3. Loaded gesicles can be collected and 4. target cells can be incubated with the gesicles in order to take up the Cas9/sgRNA complex which will lead to gene editing.
CRISPR/Cas9 gene editing in almost every cell type with less off-target effects
Since no coding gene is present in the CRISPR/Cas9 Gesicles, there is no persistent and elevated expression of Cas9. As a result, you will have reduced off-target effects after gene editing.
In addition, the CRISPR/Cas9 gesicles are engineered with nanovesicle-inducing glycoproteins on their surface that mediate binding and fusion of the gesicle with the membranes of a wide range of mammalian target cells, including harder-to-transfect cells.
