Guide-it CRISPR/Cas9 Vector Systems allow easy cloning and expression of target single guide RNAs (sgRNAs) for mammalian CRISPR/Cas9 based genome editing.
Express all components from one vector
The pGuide-it vector can simultaneously express Cas9 nuclease, a target-specific sgRNA, and an bright fluorescent protein (ZsGreen1 or tdTomato). Cas9 and the fluorescent protein are co-expressed from a bidirectional CMV promoter, and a user-defined sgRNA is cloned downstream of a human U6 promoter.
The fluorescent protein marker is ideal for monitoring transfection efficiency and/or further enriching/isolating transfected cells by FACS.
Guide-It plasmid construction is simple. First you design a complementary pair of oligos specific to your target sequence and anneal them to form a duplex. The duplexed oligos are the inserted into the supplied pre-linearized vector using the included high-efficiency ligation mix and Stellar competent cells to ensure high efficiency transformation.
