High Performance Single-Cell Transcriptome Analysis
The SMART-Seq v4 Ultra Low Input RNA Kit for the Fluidigm C1 System is an improved single-cell, full-length mRNAseq method that demonstrates high sensitivity and delivers high quality, robust and reproducible transcriptomic data.
The new chemistry employs locked nucleic acid (LNA) technology integrated in proven SMART® technology, which greatly enhances the first strand cDNA synthesis reaction. The new chemistry also utilizes efficient SeqAmp™ Polymerase, which excels even in GC- and AT-rich regions. All procedures, including lysis, RT, and all PCR steps, are designed to be performed in the Fluidigm C1 System using Open App IFC. The first generation of SMARTer Ultra Low kits for the C1 ("v1") continues to be available.
Higher sensitivity: detect more genes
The new SMART-Seq Ultra Low V4 chemistry produced more cDNA, detects significantly more genes, and achieves a significantly higher exon-to-intron mapped ratio than the older (v1) SMARTer Ultra Low chemistry.
Better detection of GC-rich genes
The components, chemistry, and protocol updates in the SMART-Seq v4 kit have also resulted in more sensitive detection of genes with high GC content.
In a direct comparison genes with high GC content - the red dots in the figure - showed higher expression with v4 than with v1.
Less technical variability - more biological relevance
cDNA libraries synthesized using the SMART-Seq V4 chemistry showed lower variability between replicates than the data generated by v1.
This reduced technical variability increases the likelihood to detect small biologically relevant differences between samples.
