Strand-specific Illumina RNA-Seq libraries from picograms of total RNA
The SMARTer Stranded Total RNA-Seq Kit - Pico Input Mammalian efficiently generates Illumina® sequencing libraries from 250 pg - 10 ng of total RNA in just 5 hours. The SMARTer cDNA synthesis method preserves the strand orientation of the original RNA, allowing you to get strand-specific sequencing data.
Works with high- and low-quality RNA (RIN 3-10)
The technologies in this product are designed to process high-quality, partially degraded, and low-quality (e.g., prepared from FFPE or LCM samples) RNA. The kit has a streamlined workflow that retains strand information and eliminates downstream library prep by incorporating indexes and adapters during the reverse-transcription and PCR-amplification steps.
Novel rRNA depletion method for small samples
Total RNA contains ~90% of ribosomal RNA sequences. Depleting these abundant rRNAs from samples before making libraries is essential to lower sequencing cost and improve mapping statistics. However, with very low input amounts, initial rRNA depletion from total RNA is not very effective and often leaves an insufficient amount of material for preparation of a good library.
This kit uses a novel technology: it removes ribosomal cDNA after the cDNA synthesis, using probes specific to mammalian ribosomal sequences. This rRNA depletion method is especially well-suited for very small samples.
